چکیده

Thrombin plays an essential role in haemostasis and thrombosis; therefore, anticoagulant strategies focus on direct or indirect inhibition of thrombin or factor Xa. Over the last 60 years, key players in antithrombotic treatment have been unfractionated heparin and vitamin K antagonists (VAKs), such as coumarin. New oral anticoagulants, with different mechanisms of action and administration, have the potential to promote dramatic changes in the management of patients with venous thromboembolic disease. This review provides an overview of the newer anticoagulants likely to be used in this indication.

An Overview of Novel Anticoagulants

Arterial and venous thromboembolism is one of the most common causes of death worldwide. For almost seventy years, heparin, low molecular weight heparin, and vitamin K antagonists have been widely used in the prevention and treatment of thromboembolic disorders. However, many of the limitations of these traditional anticoagulants have led to the search for and attempts to introduce better drug ...

New Oral Anticoagulants for the Treatment of Venous Thromboembolism: Understanding Differences and Similarities

Venous thromboembolism (VTE) is a major cause of morbidity, mortality, and healthcare expenditure. In the United States, approximately 0.1 % of the population experiences an initial VTE event each year. Anticoagulation therapy is the cornerstone of acute VTE treatment and for prevention of recurrent VTE events. Conventional anticoagulants, including heparin, low-molecular-weight heparins, fonda...

New oral anticoagulants: One Step Closer to Replacing Warfarin

Anticoagulants are widely used for the prevention and treatment of venous and arterial thrombosis. The main indications for long-term anticoagulation therapy include treatment of venous thromboembolism (VTE), stroke prevention in atrial fibrillation (AF) and the prevention of valve thrombosis in patients with mechanical heart valves. For the past 65 years, vitamin K antagonists (VKA), such as w...
